So since contact angle of the droplet seems to be the function of its volume can You please advise what droplet size I should use when testing 4 micron liquid layer? Is it correct to measure from surface to the highest point of the droplet?

From our standpoint, every research scientist measures contact angle a little bit differently as it varies from study to study. Dispense a droplet with clear angles that can be measured.
